When replacing the rear brake pads on a 2005 VW Jetta does the E-brake cable need to be disconnected in order to rotate and compress the calipers? If this is the case, is the process relatively straight forward or relatively complicated?

NO!
Use the tool to turn the piston back in, go slow, and keep the dust boot from twisting with it!
