86 Silverado- one cylinder not working. dead?

Hi. I have a 1986 chevy silverado with a fairly new 350 sbc. it has about 8k miles on it. ive been having recent problems with the holley carb i had on it so i put a edelbrock 600cfm on it. the truck actually runs pretty nicely but has the power of a 4 banger. i was checking to see if all the cylinders were firing by spraying some water on the headers. all worked fine except the 3rd one back on the passenger side(facing the front). i changed the spark plug and the old one was really nasty looking. i also changed the spark plug wire out and it still wont fire. i hope i didnt ruin the cylinder by just dumping fuel into a dead cylinder. any input would be helpful. Start with a compression test to be sure it is good. Have you replaced the plug? they do foul out if the wire is bad long enough or if you use Bosch platinum plugs
