if the transmission control module is replaced, does it need to be programmed?

yes, it does here are the insutriocn with diagrams below to help you with the repair.

Transmission Control Module Programming and Setup

1. The transmission control module (TCM) must be programmed with the proper software/calibrations. Ensure that the following conditions exist in order to prepare for TCM programming:

* The battery is fully charged.
* The ignition switch is in the RUN position.
* The Techline equipment cable connection at the data link connector (DLC) is secure.

2. Program the TCM using the latest software matching the vehicle. Refer to up-to-date Techline equipment user instructions.
3. If the TCM fails to program, proceed as follows:

1. Ensure that the TCM connection is OK.
2. Inspect the Techline equipment for the latest software version.
3. Attempt to program the TCM. If the TCM still cannot be programmed properly, replace the TCM.

Had car scanned and it said lost communication with Transmission Control Module. So I went to junkyard and got a used transmission control module from a 2006 Chevrolet Impala LT, my car is a 2006 Chevy Impala SS. They both have the same part number. So my question is, do I have to reprogram the Transmission Control Module to my vehicle or are they interchangeable?

Hello,

It looks like it does need to be programmed. In the diagrams below I have included what I found when I looked this up. You can always install it and see if it works. It wouldn't hurt any thing. If it didn't work, well then you would just have to have it programmed anyway. Let us know if we can help you with anything else.
