Intermittent Miss with Codes P0316 and P0303

This question relates to my 02 F150 XLT 2WD supercab with a 4.6 V8 Engine code W. I have a miss that appears intermittently that doesn't seem to relate to wet weather, cold, length of drive, etc. It began about 3 weeks ago when it developed the miss for 2-3 minutes without the MIL lighting. I added a bottle of Seafoam to the fuel thinking it could be water in the fuel and it did not reappear until 4-5 days ago for about a 5 minute period and the MIL lit up. The first code was P0316 followed by P0303. Since then the miss has only returned once for 30 seconds or so. Assuming that I have a COP going bad on #3 cylinder will that cause the 0316 code? Should I be testing elsewhere as well?
