Are you only having problems with the one door or all the doors when using the remote?

Drivers door is the only keyed one. Remote with new battery still doesn't work occasionally. When its locked its locked for good until finally sometime later the remote works. But the key should always work and that is what is puzzling me.

Remove the door panel and inspect the lock button rod to verify it is not binding up and inspect the harness for any damage by the window regulator. If all that looks good then the door lock assembly will need to be replaced. Also usually if you turn the key manually twice since this is a double stage lock on the second turn the other three doors should unlock as well, as well as when you use the remote. That is why I asked if the problem was only with the one door or the other locks are affected as well meaning none of the doors unlock when you press the unlock button on the remote twice?
